Semen with high number of live and normal spermatozoa is a good indicator of fertility. This study was conducted on fresh and frozen-thawed semen of water buffalo bulls used in AI to: (a) determine and compare their sperm viability; (b) determine and compare their sperm morphology; (c) determine and compare their sperm morphometry and; (d) determine the effects of cryopreservation on these sperm parameters. Five fresh and seven frozen-thawed semen samples were used in the study. A microscope with Nikon imaging software and NIS elements application installed were used to evaluate the sperm parameters. All the data gathered were reported as mean ± s.e.m and significant differences were statistically analyzed by T-test at P<0.05. Fresh semen samples have a significantly higher (p<0.05) number in live and normal spermatozoa. Sperm head on the frozen-thawed semen samples were larger. In these given results, cryopreservation greatly affects the sperm parameters in the study.
